Well, I know once you unlock it, your good. You dont have to re-unlock it everytime you wanna flash a custom rom. You would only have to relock it for the stock rom if you were gonna take in for service or something like that. Now, after that, Im kinda hesitant. I've never had the problem so I'm not to sure. Have you tried to hard reset from where it goes? Dont know if it would work but its worth a try. I would also try to flash it. Just boot it up when its hooked to your comp and run the update. See if it goes through. But honestly, Dash or Kaos will prolly know the most when it comes to this. So you should wait for one of theme for a definitive answer.
like superman said, dash or kaos would probably know best... but i think what i said earlier should work...
