how to delete a file in use?

i want to delete a file in \Windows folder, but its a file that its in use, so i cant delete it, there's a way to delete it?
using WM 6.1

my objective:
replace 2 files that will make autocorrection and autocompletation in brazilian portuguese (my language), if anyone knows another way to do it plz help.
